Abstract
A novel gene 58P1D12 and its encoded protein, and variants thereof, are described wherein 58P1D12 exhibits tissue specific expression in normal adult tissue, and is aberrantly expressed in the cancers listed in Table I. Consequently, 58P1D12 provides a diagnostic, prognostic, prophylactic and/or therapeutic target for cancer. The 58P1D12 gene or fragment thereof, or its encoded protein, or variants thereof, or a fragment thereof, can be used to elicit a humoral or cellular immune response; antibodies or T cells reactive with 58P1D12 can be used in active or passive immunization.

exact text as granted — not AI-modified1 . An isolated transcript variant of a 58P1D12 gene, wherein the transcript variant is transcribed from the 58P1D12 gene and encodes a protein comprising at least one amino acid substitution, addition or deletion relative to SEQ ID NO:12.
2 . The transcript variant of claim 1 , wherein the transcript variant comprises a nucleotide sequence selected from the group consisting of SEQ ID NO:4 or SEQ ID NO:8.
3 . The transcript variant of claim 1 , wherein the transcript variant encodes a protein comprising the amino acid sequence selected from the group consisting of SEQ ID NO:13, or SEQ ID NO:14.
4 . A recombinant expression vector comprising a transcript variant, wherein the transcript variant is transcribed from the 58P1D12 gene and encodes a protein comprising at least one amino acid substitution, addition or deletion relative to SEQ ID NO:12.
5 . The recombinant expression vector of claim 4 , wherein the transcript variant comprises a nucleotide sequence selected from the group consisting of SEQ ID NO:4 or SEQ ID NO:8.
6 . The recombinant expression vector of claim 4 , further comprising a nucleotide sequence encoding a selection marker.
7 . The recombinant expression vector of claim 4 , further comprising at least one suitable regulatory sequence operably linked to the transcript variant.
8 . A host cell comprising a recombinant expression vector comprising a transcript variant, wherein the transcript variant is transcribed from the 58P1D12 gene and encodes a protein comprising at least one amino acid substitution, addition or deletion relative to SEQ ID NO:12.
9 . The host cell of clam 8 , wherein the transcript variant comprises a nucleotide sequence selected from the group consisting of SEQ ID NO:4 or SEQ ID NO:8.
10 . The host cell of claim 8 , wherein the host cell is a prokaryotic cell.
11 . The host cell of claim 10 , wherein the prokaryotic cell is E. coli.
12 . The host cell of claim 8 , wherein the host cell is a eucaryotic cell.
13 . The host cell of claim 12 , wherein the host cell is selected from the group consisting of a yeast cell, a plant cell and an animal cell.
14 . The host cell of claim 13 , wherein the animal cell is a mammalian cell.
15 . The host cell of claim 14 , wherein the mammalian cell is selected from the group consisting of COS cells, CHO cells, 293 cells, and 293T cells.
16 . An isolated protein encoded by a transcript variant of a 58P1D12 gene, wherein the transcript variant is transcribed from the 58P1D12 gene and encodes a protein comprising at least one amino acid substitution, addition or deletion relative to SEQ ID NO:12.
17 . The protein of claim 16 , wherein the protein is encoded by a transcript comprising the nucleic acid sequence selected from the group consisting of SEQ ID NO:4, or SEQ ID NO:8.
18 . The protein of claim 16 , wherein the protein comprises the amino acid sequence selected from the group consisting of SEQ ID NO:13, or SEQ ID NO:14.
19 . An isolated antibody or fragment thereof that specifically binds to the protein of claim 16 .
20 . The antibody or fragment thereof of claim 19 , wherein the antibody is monoclonal.
21 . The antibody or fragment thereof of claim 19 , wherein the antibody is a human antibody, a humanized antibody or a chimeric antibody.
22 . The antibody or fragment thereof of claim 19 , wherein the antibody or fragment thereof is labeled with an agent.
23 . A method of inducing a mammalian immune response directed to a protein encoded by a transcript variant of a 58P1D12 gene, wherein the transcript variant is transcribed from the 58P1D12 gene and encodes a protein comprising at least one amino acid substitution, addition or deletion relative to SEQ ID NO:12, comprising:
exposing cells of the immune system of a mammal to an immunogenic portion of the protein comprising at least one amino acid substitution, addition or deletion relative to SEQ ID NO:12; or the transcript variant, or a combination of both, whereby the mammalian immune response directed to the protein is induced.
24 . The method of claim 23 , wherein the immunogenic portion comprises at least one T cell or at least one B cell epitope.
25 . The method of claim 24 , whereby the immunogenic portion induces a B cell to generate antibodies that specifically bind to the protein.
26 . The method of claim 24 , whereby the immunogenic portion activates a cytotoxic T cell (CTL) that kills an autologous cell that expresses the protein.
27 . The method of claim 24 , wherein the T cell is a helper T cell (HTL), whereby the immunogenic portion activates an HTL that secretes cytokines that facilitate the cytotoxic activity of a CTL or the antibody producing activity of a B cell.
28 . The method of claim 23 , wherein the immune response comprises production of an antibody that binds specifically to the protein.
29 . The method of claim 23 , wherein the protein is encoded by a transcript variant comprising the nucleic acid sequence selected from the group consisting of SEQ ID NO:4, or SEQ ID NO:8.
30 . The method of claim 23 , wherein the protein comprises the amino acid sequence selected from the group consisting of SEQ ID NO:13, or SEQ ID NO:14.
31 . An immunogenic composition comprising an immunogenic portion of:
a protein encoded by a transcript variant of a 58P1D12 gene, wherein the transcript variant is transcribed from the 58P1D12 gene and encodes a protein comprising at least one amino acid substitution, addition or deletion relative to SEQ ID NO:12; and/or transcript variant that encodes the protein.
32 . The immunogenic composition of claim 31 , wherein the protein is encoded by a transcript variant comprising the nucleic acid sequence selected from the group consisting of SEQ ID NO:4, or SEQ ID NO:8.
33 . The immunogenic composition of claim 31 , wherein the protein comprises the amino acid sequence selected from the group consisting of SEQ ID NO:13, or SEQ ID NO:14.
