Abstract
The present invention relates to morpholinyl, and pyrrolidinyl analogs, pharmaceutical compositions containing them, and their use as antagonists of urotensin II.

A is (CH 2 ) n ;
n is 1 or 2;
the group (a) may be unsubstituted or substituted by one, two, or three substituents chosen from: halo, OC 1-3 alkyl, OH, CN, C(O)OH, NR 7 R 8 , OCF 3 , C(O)NR 9 R 10 , SO 2 C 1-3 alkyl, SO 2 C 3-6 cycloalkyl, SO 2 -phenyl, SO 2 NR 9 R 10 , CF 3 , and NO 2 ;
R 2 is hydrogen, halo, OC 1-3 alkyl, NHC(O)C 1-3 alkyl, NHSO 2 C 1-3 alkyl, NHSO 2 -phenyl, NHC(O)C 3-6 cycloalkyl, or NHC(O)-phenyl;
R 3 is C 1-3 alkyl or C 3-6 cycloalkyl;
R 4 , R 5 , and R 6 are independently hydrogen or CH 3 ;
R 7 and R 8 are independently hydrogen, C(O)C 1-3 alkyl, C(O)C 3-6 cycloalkyl, C(O)-phenyl, C 1-6 alkyl, phenyl, SO 2 C 1-3 alkyl, SO 2 C 3-6 cycloalkyl, SO 2 -phenyl, or C(O)NHC 1-3 alkyl;
R 9 and R 10 are independently hydrogen, C 1-3 alkyl, C 3-6 cycloalkyl, phenyl;
or taken together R 9 and R 10 may form a 5 to 7 member ring optionally containing one or two heteroatoms chosen from O, N, and S;
X is O or a bond;
B is:
R 12 is hydrogen, C 1-3 alkyl, CH 2 CN, (CH 2 ) m (O)CH 3 , CH 2 C(O)NH 2 , or (CH 2 ) m OH;
R 13 is hydrogen, halo, C 1-3 alkyl, CF 3 , or OC 1-3 alkyl;
m is 2 or 3;
R 14 is hydrogen, halo, C 1-3 alkyl, OC 1-3 alkyl, OCF 3 , CF 3 , C(O)NR 15 R 16 , or C(O)OC 1-3 alkyl; and
R 15 and R 16 are independently hydrogen, C 1-3 alkyl, C 3-6 cycloalkyl, or phenyl;
or a pharmaceutically acceptable salt thereof.
2 . A compound of claim 1 wherein:
R 1 is phenyl substituted by one or two halo, OC 1-3 alkyl, CN, C(O)OH, NR 7 R 8 , C(O)NR 9 R 10 , SO 2 C 1-3 alkyl, or SO 2 NR 11 R 12 ; R 2 is hydrogen or halo; R 3 is methyl; R 4 is hydrogen or CH 3 ; R 5 and R 6 are hydrogen; R 7 and R 8 are independently hydrogen, C(O)C 1-3 alkyl, C 1-6 alkyl, SO 2 C 1-3 alkyl, or C(O)NHC 1-3 alkyl; R 9 and R 10 are independently H, C 1-3 alkyl; or taken together R 9 and R 10 form a pyrrolidine, morpholine, or piperidine group; X is O or a bond; B is:
R 12 is methyl, CH 2 CN, (CH 2 ) m (O)CH 3 , or CH 2 C(O)NH 2 ;
R 13 is hydrogen or halo;
m is 2; and
R 14 is hydrogen or halo;
or a pharmaceutically acceptable salt thereof.
3 . A compound of claims 1 and 2 wherein:
R 1 is phenyl substituted by one or two OC 1-3 alkyl, C(O)OH, or C(O)NR 9 R 10 ; R 2 is hydrogen or halo; R 3 is methyl; R 4 is hydrogen or CH 3 ; R 5 and R 6 are hydrogen; R 9 and R 10 are independently hydrogen, C 1-3 alkyl; or taken together R 9 and R 10 form a pyrrolidine, morpholine, or piperidine group; X is O or a bond; B is:
