Abstract

The invention provides a palindromic immune modulatory nucleic acid. The invention also provides methods for generating, enhancing and modifying the immune response caused by palindromic immune modulatory compounds used for immunotherapy applications

A method for therapeutically treating a vertebrate having cancer, an autoimmune disorder, airway inflammation, inflammatory disorders, skin disorders, allergy, asthma or a disease caused by a pathogen, such method comprising administering to the patient a palindromic immune modulatory oligonucleotide having a structure selected from 5′-TCG 1 TCG 1 TTCTC-Y-G 1 CTTG 1 CAAG 1 CT-5′, 5′-TCG 1 TCG 1 TTCTC-Y-GAG 1 CTTG 1 CAAG 1 CTCT-5′, 5′-TCG 1 TCG 1 TTCTC-Y-GAGAG 1 CTTG 1 CAAG 1 CTCTCT-5′, 5′-TCG 1 TCG 1 TTCTC-Y-GCCGCGCGGC-5′, 5′-TCG 1 TCG 1 TTAGA-Y-TG 1 CTG 1 CT-5′, or 5′-TCG 1 TCG 1 TTC-Y-G 1 CTTG 1 CAAG 1 CT-5′; wherein X is a glycerol linker, Y is C3-linker and G 1  is 7-deazaguanosine.

The method according to  claim 2 , wherein the route of administration is selected from parenteral, oral, sublingual, transdermal, topical, mucosal, inhalation, intranasal, aerosol, intraocular, intratracheal, intrarectal, vaginal, gene gun, dermal patch, eye drop and mouthwash.

A method for preventing cancer, an autoimmune disorder, airway inflammation, inflammatory disorders, skin disorders, allergy, asthma or a disease caused by a pathogen in a vertebrate, such method comprising administering to the vertebrate a palindromic immune modulatory oligonucleotide having a structure selected from 5′-TCG 1 TCG 1 TTCTC-Y-G 1 CTTG 1 CAAG 1 CT-5′, 5′-TCG 1 TCG 1 TTCTC-Y-GAG 1 CTTG 1 CAAG 1 CTCT-5′, 5′-TCG 1 TCG 1 TTCTC-Y-GAGAG 1 CTTG 1 CAAG 1 CTCTCT-5′, 5′-TCG 1 TCG 1 TTCTC-Y-GCCGCGCGGC-5′, 5′-TCG 1 TCG 1 TTAGA-Y-TG 1 CTG 1 CT-5′, or 5′-TCG 1 TCG 1 TTC-Y-G 1 CTTG 1 CAAG 1 CT-5′; wherein X is a glycerol linker, Y is C3-linker and G 1  is 7-deazaguanosine.
