Abstract
The present invention relates to novel oxadiazole derivatives having pharmacological activity, processes for their preparation, pharmaceutical compositions containing them and their use in the treatment of various disorders.

exact text as granted — not AI-modified1 . A compound of formula (I) or a pharmaceutically acceptable salt thereof:
A is phenyl or a 5 or 6-membered heteroaryl ring;
R 1 is up to two substituents independently selected from halogen, C (1-3) alkoxy, C (1-3) fluoroalkyl, cyano, optionally substituted phenyl, C (1-3) fluoroalkoxy, C (1-6) alkyl and C (3-6) cycloalkyl;
R 2 is hydrogen, halogen or C (1-4) alkyl;
B is a 7 membered saturated ring selected from the following:
R 3 is hydrogen or (CH 2 ) 1-4 CO 2 H;
R 4 is hydrogen or C (1-3) alkyl optionally interrupted by oxygen;
2 . A compound of formula (I) or a pharmaceutically acceptable salt thereof, wherein:
A is phenyl; R 1 is up to two substituents independently selected from chloro, isopropoxy and cyano; R 2 is hydrogen; B is (a); R 3 is hydrogen or (CH 2 ) 1-3 CO 2 H; R 4 is hydrogen.
3 . A compound of formula (I) or a pharmaceutically acceptable salt thereof, wherein:
A is phenyl or pyridinyl; and/or R 1 is up to two substituents independently selected from chloro, isopropoxy and cyano; and/or R 2 is hydrogen; and/or B is (b); and/or R 3 is hydrogen or (CH 2 ) 1-4 CO 2 H; and/or R 4 is hydrogen.
4 . A compound of formula (I) or a pharmaceutically acceptable salt thereof, wherein:
A is phenyl or pyridinyl; R 1 is up to two substituents independently selected from chloro, methoxy, isopropoxy, trifluoromethyl, phenyl and cyano; R 2 is hydrogen; B is (c); R 3 is hydrogen or (CH 2 ) 1-3 CO 2 H; R 4 is hydrogen.
5 . A compound according to claim 1 selected from:

[7-(5-{3-chloro-4-[(1-methylethyl)oxy]phenyl}-1,2,4-oxadiazol-3-yl)-1,3,4,5-tetrahydro-2H-2-benzazepin-2-yl]acetic acid 2-[(1-methylethyl)oxy]-5-[3-(2,3,4,5-tetrahydro-1H-2-benzazepin-7-yl)-1,2,4-oxadiazol-5-yl]benzonitrile 6 . A compound according to claim 1 , which is
4-[7-(5-{3-cyano-4-[(1-methylethyl)oxy]phenyl}-1,2,4-oxadiazol-3-yl)-1,2,4,5-tetrahydro-3H-3-benzazepin-3-yl]butanoic acid hydrochloride or a pharmaceutically acceptable salt thereof.
7 . A compound according to claim 1 , which is
4-[9-(5-{3-Chloro-4-[(1-methylethyl)oxy]phenyl}-1,2,4-oxadiazol-3-yl)-2,3-dihydro-1,4-benzoxazepin-4(5H)-yl]butanoic acid or a pharmaceutically acceptable salt thereof.
8 . Use of a compound according to any one of claims 1 to 7 for the treatment of conditions or disorders mediated by S1P1 receptors.

Use according to claim 8 , wherein the condition or disorder is multiple sclerosis, autoimmune diseases, chronic inflammatory disorders, asthma, inflammatory neuropathies, arthritis, transplantation, Crohn's disease, ulcerative colitis, lupus erythematosis, psoriasis, ischemia-reperfusion injury, solid tumours, and tumour metastasis, diseases associated with angiogenesis, vascular diseases, pain conditions, acute viral diseases, inflammatory bowel conditions, insulin and non-insulin dependant diabetes.
10 . Use according to claim 9 , wherein the condition is lupus erythematosis.
11 . Use of a compound according to any one of claims 1 to 7 to manufacture a medicament for use in the treatment of conditions or disorders mediated by S1P1 receptors.

Use according to claim 11 , wherein the condition or disorder multiple sclerosis, autoimmune diseases, chronic inflammatory disorders, asthma, inflammatory neuropathies, arthritis, transplantation, Crohn's disease, ulcerative colitis, lupus erythematosis, psoriasis, ischemia-reperfusion injury, solid tumours, and tumour metastasis, diseases associated with angiogenesis, vascular diseases, pain conditions, acute viral diseases, inflammatory bowel conditions, insulin and non-insulin dependant diabetes.
13 . Use according to claim 12 , wherein the condition is lupus erythematosis.
14 . A pharmaceutical composition comprising a compound according to any one of claims 1 to 7 .
15 . A method of treatment for conditions or disorders in mammals including humans which can be mediated via the S1P1 receptors which comprises administering to the sufferer a therapeutically safe and effective amount of a compound of formula (I) or a pharmaceutically acceptable salt thereof.
