Nucleotide sequence encoding a modulator of nf-kb
Abstract
The present invention relates to nucleotide sequences encoding a modulator of NF-κB, and to the polypeptides encoded by the nucleotide sequences. In particular, the invention relates to nucleotide sequences and the polypeptides encoded thereby, wherein the polypeptides are involved in the response to NF-κB-activating stimuli, including HTLV-1 Tax, LPS, PMA and IL-1. The invention also relates to antibodies to the modulator of NF-κB, methods of detecting modulator of NF-κB using the antibodies, methods of treatment associated with NF-κB activation and to methods of identifying compounds which modulate the activity of the modulator of NF-κB.

51 . A purified modulator of NF-κB comprising a polypeptide having an amino acid sequence that is at least 95% homologous to SEQ ID NO: 1, or an active fragment thereof, wherein the purified modulator of NF-κB possesses the following structural and functional characteristics:
a) an apparent molecular weight of approximately 48 kD; b) a pI of approximately 5.66; c) containing a leucine zipper motif; and d) binding to a kinase involved in the activation of NF-κB.
52 . The purified modulator of NF-κB according to claim 51 , wherein the purified modulator of NF-κB is labeled with a detectable label.
53 . The purified modulator of NF-κB according to claim 52 , wherein the detectable label is selected from the group consisting of fluorescent labels, radioactive labels, and enzymatic labels.
54 . The purified modulator of NF-κB according to claim 53 , wherein the detectable label is one or more fluorescent labels selected from the group consisting of fluorescein, rhodamine, auramine, Texas Red, AMCA Blue, and Lucifer Yellow.
55 . The purified modulator of NF-κB according to claim 53 , wherein the detectable label is one or more radioactive labels selected from the group consisting of 3 H, 14 C, 32 P, 35 S, 36 Cl, 51 Cr, 57 Co, 58 Co, 59 Fe, 90 Y, 125 I, 131 I, and 186 Re.
56 . The purified modulator of NF-κB according to claim 53 , wherein the detectable label is one or more enzymatic labels selected from the group consisting of peroxidase, β-glucuronidase, β-D-glucosidase, β-D-galactosidase, urease, glucose oxidase, and alkaline phosphatase.
57 . A composition comprising the purified modulator of NF-κB according to claim 51 , and a pharmaceutically acceptable carrier.
58 . The composition according to claim 57 , wherein the pharmaceutically acceptable carrier is one or more excipients selected from the group consisting of water, saline, dextrose, glycerol, and ethanol.
59 . A purified modulator of NF-κB comprising a polypeptide encoded by a polynucleotide sequence that is at least 95% homologous to SEQ ID NO: 2, or an active fragment thereof, wherein the purified modulator of NF-κB possesses the following structural and functional characteristics:
a) an apparent molecular weight of approximately 48 kD; b) a pI of approximately 5.66; c) containing a leucine zipper motif; and d) binding to a kinase involved in the activation of NF-κB.
60 . The purified modulator of NF-κB according to claim 59 , wherein the purified modulator of NF-κB is labeled with a detectable label.
61 . The purified modulator of NF-κB according to claim 60 , wherein the detectable label is selected from the group consisting of fluorescent labels, radioactive labels, and enzymatic labels.
