Abstract
The present invention is directed to methods for delivering stem cells to a target tissue in a mammal using glycoconjugate to traffic the stem cells to a desired organ in the mammal. The methods according to the present invention are especially applicable to administering stem cells such as those derived from the bone marrow or from umbilical cord tissue. The methods are also useful for targeting a gene of interest to a tissue in a mammal by introducing a cell containing the gene of interest and administering a glycoconjugate to the mammal.

A method for directing stem cells to a target tissue on a subject, comprising administering to the subject (i) a composition comprising an asialoglycoprotein receptor blocker, and (ii)(a) when the target tissue is heart tissue, a composition comprising hematopoietic stem cells, or (ii)(b) when the target tissue is liver tissue, a composition comprising mesenchymal stem cells.
2 . A method according to claim 1 wherein the stem cells are CD34 + stem cells.
3 . A method according to claim 1 wherein when the target tissue is heart tissue, the hematopoietic stem cells are CD34 + hematopoietic stem cells, and when the target tissue is liver tissue, the mesenchymal stem cells are selected from the group consisting of stromal cells from bone marrow, stromals cell from liposuction fat, and stromal progenitor cells from a cord blood-depleted expelled placenta.
4 . A method according to claim 1 wherein the asialoglycoprotein receptor blocker is asialoorosomucoid.

A method according to claim 1 wherein the asialoglycoprotein receptor blocker is selected from the group consisting of asialofetuin, galactose-bonded polylysine, galactose-bonded polyglucoseamine, and a glycoprotein comprising a carbohydrate structure selected from the group consisting of carbohydrate structures A, B, C, and D in FIG. 3 .
6 . A method according to claim 5 wherein the glycoprotein is a human serum glycoprotein.
7 . A method according to claim 1 wherein the composition comprising the asialoglycoprotein receptor blocker is administered before the composition comprising the stem cells is administered.
8 . A method according to claim 1 wherein the composition comprising the asialoglycoprotein receptor blocker is administered before and after the composition comprising the stem cells is administered.
9 . A method according to claim 1 wherein the composition comprising the asialoglycoprotein receptor blocker is administered during administration of the composition comprising the stem cells.
10 . A method according to claim 1 wherein the subject is a human subject.
11 . A method according to claim 1 wherein the stem cells comprise a therapeutic gene.
12 . A method for directing asialodeterminant-bearing stem cells to a target tissue in a subject, comprising administering to the subject (i) a composition comprising an asialoglycoprotein receptor blocker, and (ii)(a) when the target tissue is heart tissue, a composition comprising asialodeterminant-bearing stem cells, wherein the asialodeterminant-bearing stem cells are not mesenchymal stem cells but are optionally hematopoietic stem cells or (ii)(b) when the target tissue is liver tissue, a composition comprising asialodeterminant-bearing stem cells, wherein the asialodeterminant-bearing stem cells are not hematopoietic stem cells but are optionally mesenchymal stem cells.
13 . A method according to claim 11 wherein the asialoglycoprotein receptor blocker is asialoorosomucoid.
14 . A method according to claim 11 wherein the asialoglycoprotein receptor blocker is selected from the group consisting of asialofetuin, a galactose-bonded polylysine, a galactose-bonded polyglucoseamine, and a glycoprotein comprising a carbohydrate structure selected from the group consisting of carbohydrate structures A, B, C, and D in FIG. 3 .
15 . A method according to claim 13 wherein the glycoprotein is a human serum glycoprotein.
16 . A method according to claim 11 wherein the composition comprising the asialoglycoprotein receptor blocker is administered before the composition comprising the stem cells is administered.
17 . A method according to claim 11 wherein the composition comprising the asialoglycoprotein receptor blocker is administered before and after the composition comprising the stem cells is administered.
18 . A method according to claim 11 wherein the composition comprising the asialoglycoprotein receptor blocker is administered during administration of the composition comprising the stem cells.
19 . A method according to claim 11 wherein the subject is a human subject.
