Abstract

The present invention relates to variants (mutants) of parent Termamyl-like alpha-amylases, which variant has alpha-amylase activity and exhibits altered stability, in particular at high temperatures and/or at low pH relative, and/or low Ca 2+ to the parent alpha-amylase.

         27 . A variant of a parent alpha-amylase, comprising a substitution of the amino acid at position 239, wherein
 (a) the parent alpha-amylase has at least 90% sequence identity to SEQ ID NO: 8,   (b) the variant has alpha-amylase activity, and   (c) the variant has 1-15 mutations relative to the parent alpha-amylase, and   (d) each position corresponds to a position of the amino acid sequence of SEQ ID NO: 8.   
     
     
         28 . The variant of  claim 27 , wherein the parent alpha-amylase has at least 95% identity to SEQ ID NO: 8. 
     
     
         29 . The variant of  claim 27 , wherein the parent alpha-amylase has at least 97% identity to SEQ ID NO: 8. 
     
     
         30 . The variant of  claim 27 , wherein the parent alpha-amylase has at least 99% identity to SEQ ID NO: 8. 
     
     
         31 . The variant of  claim 27 , wherein the parent alpha-amylase comprises SEQ ID NO: 8. 
     
     
         32 . The variant of  claim 27 , wherein the 1-15 mutations are substitutions. 
     
     
         33 . The variant of  claim 27 , wherein the substitution of the amino acid at position 239 is the only mutation. 
     
     
         34 . The variant of  claim 27 , which contains two mutations. 
     
     
         35 . The variant of  claim 27 , which contains three, four or five mutations.

         37 . A composition comprising an alpha-amylase variant of  claim 27  and one or more enzymes selected from the group consisting of glucoamylase, phytase and pullulanase.

         39 . A variant of a parent alpha-amylase, comprising a substitution of the amino acid at position 239, wherein
 (a) the parent alpha-amylase has at least 90% sequence identity to SEQ ID NO: 6,   (b) the variant has alpha-amylase activity, and   (c) the variant has 1-15 mutations relative to the parent alpha-amylase, and   (d) each position corresponds to a position of the amino acid sequence of SEQ ID NO: 8.   
     
     
         40 . The variant of  claim 39 , wherein the parent alpha-amylase has at least 95% identity to SEQ ID NO: 6. 
     
     
         41 . The variant of  claim 39 , wherein the parent alpha-amylase has at least 97% identity to SEQ ID NO: 6. 
     
     
         42 . The variant of  claim 39 , wherein the parent alpha-amylase has at least 99% identity to SEQ ID NO: 6. 
     
     
         43 . The variant of  claim 39 , wherein the parent alpha-amylase comprises SEQ ID NO: 6.
