Process and intermediates for the preparation of 3-(amino)-3-cyclobutylmethyl-2-hydroxy-propionamide or salts thereof

Abstract

In one embodiment, the present application relates to a process of making a compound of formula I: and to certain intermediate compounds that are made within the process of making the compound of formula I.

1 . A process for the preparation of a precipitate comprising predominantly either the RS or SR enantiomer of Formula VIII; 
     
       
         
         
             
             
         
       
       the process comprising: 
       (i) extracting the isolated supernatant liquid present in Step “c” of claim  25  with an acetate to obtain an acetate solution comprising RS and SR compounds of Formula VIII; and 
       (ii) forming a precipitate from the acetate solution comprising predominately the RS and SR compounds of Formula VIII. 
     
   
   
       2 . A process for preparing a reaction mixture precipitate comprising predominantly either the RR or SS enantiomer of Formula VIII; 
     
       
         
         
             
             
         
       
       the process comprising: 
       (a) providing a reaction mixture by treating a DMSO solution of the compound of VII, 
     
     
       
         
         
             
             
         
       
        with a base in a suitable solvent and subsequently reacting it with peroxide; and 
       (b) precipitating the compounds of VIII RR and SS by adding an anti-solvent to the reaction mixture provided by step “a”. 
     
   
   
       3 . The process of  claim 2  wherein the base solvent in step “a” is acetone and the anti-solvent in step “b” is water.
