Hydroxy Acid Complexes for Antiaging and Skin Renovation
Abstract
This invention relates to certain divalent or polyvalent metals of first transition series of the Periodic Table of Elements and including zinc and Molybdenum, complexed concurrently with an amino acid and a hydroxy acid in a novel spirocyclic bidentate chemical complex form (FIG. 1 ). Upon topical application said metal complexes undergo enhanced skin penetration without causing skin irritation. The said metal complexes are useful for topical conditions that includes dry skin, xerosis, ichthyosis, dandruff, brownish spots, keratoses, melasma, lentigines, age spots, dark circles around eyes, skin pigmentation, topical inflammation, liver spots, pigmented spots, wrinkles, blemishes, skin lines, oily skin, acne, warts, eczema, pruritic skin, psoriasis, inflammatory dermatoses, disturbed keratinization, dandruff, bacterial infection, fungal infection, wound healing, body odor, and skin changes associated with aging:

Wherein a metal [M] is complexed with an amino acid [AA], which is also complexed with a hydroxy acid [HA] at the same metal; and, wherein,
M is selected from the group consisting of Ti, V, Cr, Mn, Fe, Co, Ni, Cu, Zn, and Mo; and,

AA is selected from the group consisting of Threonine, tyrosine, cysteine, methionine, aspartic acid, asparagine, glutamic acid, glutamine, arginine, lysine, histidine, phenylalanine, tryptophane, proline, hydroxyproline, beta-alanine, beta-aminoisobutanoic acid, homocysteine, homoserine, ornithine, citrulline, 5-amino levulinic acid, glycine, and picolinic acid.
2 . A composition comprising the compound of claim 1 , wherein said composition causes the treatment of dermatological disorders.
3 . A composition comprising the compound of claim 1 , wherein said compound is zinc glycinate salicylate.

A composition comprising the compound of claim 1 , wherein said compound is selected from the group consisting of zinc glycinate salicylate, zinc glycinate glycolate, zinc glycinate lactate, zinc glycinate mandelate, zinc glycinate malate, zinc glycinate hydroxycitrate, zinc glycinate gluconate, Manganese glycinate salicylate, manganese glycinate glycolate, manganese glycinate lactate, manganese glycinate mandelate, manganese glycinate malate, manganese glycinate hydroxycitrate, Manganese glycinate gluconate, Copper glycinate salicylate, copper glycinate glycolate, copper glycinate lactate, copper glycinate mandelate, copper glycinate malate, copper glycinate gluconate, copper glycinate hydroxycitrate, and combinations thereof.

A composition according to claim 2 , wherein said dermatological disorder is selected from the group consisting of dry skin, xerosis, ichthyosis, dandruff, brownish spots, keratoses, melasma, lentigines, age spots, liver spots, pigmented spots, dark circles under the eyes, skin pigmentation, dark skin, skin wrinkles, blemishes, skin lines, oily skin, acne, warts, eczema, pruritic skin, psoriasis, inflammatory dermatoses, topical inflammation, disturbed keratinization, skin changes associated with aging, nail or skin requiring cleansers, conditioning or treatment, and hair or scalp requiring shampooing or conditioning, and combinations thereof.
6 . A composition comprising the compound of claim 1 , wherein said compound is zinc glycinate glycolate.
7 . A composition comprising the compound of claim 1 , wherein said compound is zinc glycinate hydroxycitrate.
8 . A composition comprising the compound of claim 1 , wherein said composition is for topical application.
9 . A composition according to claim 2 , and a carrier or base.
10 . A composition according to claim 2 , and a pH more than or equal to 3.5 but less than or equal to 7.5.
11 . A composition according to claim 4 , wherein said compound is zinc glycinate salicylate.
12 . A composition according to claim 5 , wherein said dermatological disorder is dandruff.
13 . A composition according to claim 5 , wherein said dermatological disorder is skin changes associated with aging, including wrinkles.
14 . A composition according to claim 5 , wherein said dermatological disorder is skin pigmentation, including dark circles around eyes.
15 . A composition according to claim 5 , wherein said dermatological disorder is skin lines.
16 . A composition according to claim 5 , wherein said dermatological disorder is acne.
17 . A composition according to claim 5 , wherein said dermatological disorder is dark skin.

A process for metal complex of formula (I), and comprising; mixing of (i) a hydroxy acid, and (ii) a metal complex of an amino acid, and (iii) a solubilizer, and, (iv) wherein (i) and (ii) are present in nearly equimolar weight percent amounts, and (v) wherein (i) and (ii) undergo a complexation reaction to form said metal complex, and wherein said metal complex having spirocyclic chemical structure of formula (I).

A process for metal complex of formula (I) according to claim 18 , and comprising; the mixing of (i) a metal salt of a hydroxy acid, and (ii) a metal complex of an amino acid, and (iii) a solubilizer, and (iv) wherein (i) and (ii) are present in nearly equimolar weight percent amounts, and (v) wherein (i) and (ii) undergo an intermolecular complexation reaction to form said metal complex, and wherein said metal complexes having general spirocyclic chemical structures of formula (I).

A process for a metal complex of hydroxy acids, and comprising; the mixing of (i) a metal complex of one hydroxy acid, and (ii) a metal complex of a second hydroxy acid, and (iii) a solubilizer, and (iv) wherein (i) and (ii) are present in nearly equimolar weight percent amounts, and (v) wherein (i) and (ii) undergo an intermolecular complexation reaction to form a metal complex of two different hydroxy acids and wherein said metal complexes having general spirocyclic chemical structures formula (II);

M is selected from the group consisting of Ti, V, Cr, Mn, Fe, Co, Ni, Cu. Zn, and Mo; and,

21 . A method of treating aging related skin conditions comprising topically applying to the skin, for a period of time and in an amount sufficient to effect changes in the dermis, of a composition comprising the compound of claim 1 .
22 . A method according to claim 21 , wherein said method and a pH more than or equal to 3.5 but less than or equal to 7.5.
23 . A method according to claim 21 , wherein said method causes the reduction of pigmented and non-pigmented age spots.
24 . A method according to claim 21 , wherein said method causes a substantial increase in skin thickness.
25 . A method according to claim 21 , wherein said method stimulates synthesis of a dermal component selected from the group consisting of glycosaminoglycans, proteoglycans, collagen and elastic fibers.
26 . A method according to claim 21 , wherein said method reduces dandruff.
27 . A method according to claim 21 , wherein said method reduces bacterial infection.
28 . A method according to claim 21 , wherein said method reduces fungal infection.
29 . A method according to claim 21 , wherein said method accelerates wound healing.
