Abstract

Monoclonal antibodies, and antigen binding fragments thereof, which bind to Protein A of Staphylococcus aureus are provided.

1 . An isolated polynucleotide comprising a coding sequence for an antigen-binding polypeptide;
 wherein the antigen-binding polypeptide binds to the same epitope of Protein A as monoclonal antibody 76 produced by hybridoma cell line 358A76.1 as deposited with the American Type Culture Collection and assigned accession number PTA-7938;   wherein the isolated polynucleotide comprises a nucleotide sequence that has at least 75% sequence identity to a nucleotide sequence encoding the monoclonal antibody 76.   
     
     
         2 . The isolated polynucleotide of  claim 1 , wherein the coding sequence hybridizes under high stringency to the gene in hybridoma cell line 358A76.1 encoding monoclonal antibody 76. 
     
     
         3 . An isolated polynucleotide comprising the nucleic acid sequence coding for the heavy chain, the light chain, the heavy chain variable region, the light chain variable region, or one or more complementarity determining regions of monoclonal antibody 76 produced by hybridoma cell line 358A76.1 as deposited with the American Type Culture Collection and assigned accession number PTA-7938. 
     
     
         4 . The isolated polynucleotide of  claim 3 , comprising the nucleic acid sequence coding for the heavy chain. 
     
     
         5 . The isolated polynucleotide of  claim 4 , comprising the nucleic acid sequence coding for the heavy chain variable region. 
     
     
         6 . The isolated polynucleotide of  claim 3 , comprising the nucleic acid sequence coding for the light chain. 
     
     
         7 . The isolated polynucleotide of  claim 6 , comprising the nucleic acid sequence coding for the light chain variable region. 
     
     
         8 . A recombinant vector comprising the nucleic acid sequence of the isolated polynucleotide of any one of  claims 1 - 3 . 
     
     
         9 . The recombinant vector of  claim 8 , wherein the recombinant vector is an expression vector. 
     
     
         10 . A host cell comprising the recombinant vector of  claim 8 . 
     
     
         11 . A method of producing a substantially purified antibody, or antigen binding fragment thereof, the method comprising growing a host cell of  claim 10  under conditions in which the antibody, or antigen binding fragment thereof, is expressed and harvesting the expressed antibody, or antigen binding fragment thereof.
