Abstract

The present invention provides peptides libraries which are useful for rapid identification of biologically active compounds. The invention further provides peptides which include cell-growth affecting peptides and peptides which enhance or inhibit production of cellular proteins. Many of the peptides of the invention may be produced in large quantity by recombinant techniques and formulated in culture medium to produce the desired effect on cultured cells and tissues. Certain of the libraries of the invention and the peptides identified in them are particularly useful in concatemer-based recombinant expression methods.

A cell or tissue culture medium comprising a peptide consisting of an amino acid sequence selected from the group consisting of HTVEL (SEQ ID NO:26), GGMPG (SEQ ID NO:45), SYIME (SEQ ID NO:46), YEYIY (SEQ ID NO:47), GHPVE (SEQ ID NO:50), LSFIE (SEQ ID NO:63), FEFGV (SEQ ID NO:64), GDYVS (SEQ ID NO:65), ETVNF (SEQ ID NO:66), and YEYIG (SEQ ID NO:69). 
     
     
         2 . The culture medium of  claim 1  which is a chemically defined medium, a serum-free medium or a hydrolysate-free medium. 
     
     
         3 . The culture medium of  claim 1  which comprises about 0.1-25 mM of the peptide. 
     
     
         4 . The culture medium of  claim 3  which comprises about 1.0-12 mM of the peptide. 
     
     
         5 . The cell or tissue culture medium of  claim 1 , wherein said peptide is HTVEL (SEQ ID NO:26). 
     
     
         6 . The cell or tissue culture medium of  claim 1 , wherein said peptide is GGMPG (SEQ ID NO:45). 
     
     
         7 . The cell or tissue culture medium of  claim 1 , wherein said peptide is SYIME (SEQ ID NO:46). 
     
     
         8 . The cell or tissue culture medium of  claim 1 , wherein said peptide is YEYIY (SEQ ID NO:47). 
     
     
         9 . The cell or tissue culture medium of  claim 1 , wherein said peptide is GHPVE (SEQ ID NO:50). 
     
     
         10 . The cell or tissue culture medium of  claim 1 , wherein said peptide is LSFIE (SEQ ID NO:63). 
     
     
         11 . The cell or tissue culture medium of  claim 1 , wherein said peptide is FEFGV (SEQ ID NO:64). 
     
     
         12 . The cell or tissue culture medium of  claim 1 , wherein said peptide is GDYVS (SEQ ID NO:65). 
     
     
         13 . The cell or tissue culture medium of  claim 1 , wherein said peptide is ETVNF (SEQ ID NO:66). 
     
     
         14 . The cell or tissue culture medium of  claim 1 , wherein said peptide is YEYIG (SEQ ID NO:69).

A method for enhancing cellular protein production comprising culturing cells or tissues in the presence of a peptide having an amino acid sequence consisting of an amino acid sequence selected from the group consisting of HTVEL (SEQ ID NO:26), GGMPG (SEQ ID NO:45), SYIME (SEQ ID NO:46), YEYIY (SEQ ID NO:47), GHPVE (SEQ ID NO:50), LSFIE (SEQ ID NO:63), FEFGV (SEQ ID NO:64), GDYVS (SEQ ID NO:65), ETVNF (SEQ ID NO:66), and YEYIG (SEQ ID NO:69). 
     
     
         16 . The method of  claim 15  wherein production of β-toxin is enhanced. 
     
     
         17 . The method of  claim 15  wherein the cells are cultured in the presence of about 0.1-25 mM of the peptide. 
     
     
         18 . The method of  claim 17  wherein the cells are cultured in the presence of about 1.0-12 mM of the peptide. 
     
     
         19 . The method of  claim 15 , wherein said peptide is HTVEL (SEQ ID NO:26). 
     
     
         20 . The method of  claim 15 , wherein said peptide is GGMPG (SEQ ID NO:45). 
     
     
         21 . The method of  claim 15 , wherein said peptide is SYIME (SEQ ID NO:46). 
     
     
         22 . The method of  claim 15 , wherein said peptide is YEYIY (SEQ ID NO:47). 
     
     
         23 . The method of  claim 15 , wherein said peptide is GHPVE (SEQ ID NO:50).
