Abstract
This invention relates to novel lactams having the formula (I): to their pharmaceutical compositions and to their methods of use. These novel compounds inhibit the processing of amyloid precursor protein and, more specifically, inhibit the production of Aβ-peptide, thereby acting to prevent the formation of neurological deposits of amyloid protein. More particularly, the present invention relates to the treatment of neurological disorders related to β-amyloid production such as Alzheimer's disease and Down's Syndrome.

8 . A process according to claim 1 , for preparing a compound of formula (Ie)
or a stereoisomer or a pharmaceutically acceptable salt form thereof, wherein
Q is n-butyl, n-pentyl, n-hexyl, 2-(furan-2-yl)-ethyl, and Z-Y—X—W— is methyl, Q is n-butyl when Z-Y—X—W is 2-diethylaminoethyl, 3-hydroxypropyl, n-butyl or benzyl.
9 . A method for preparing a pharmaceutical composition comprising combining a compound prepared according to the process of claim 1 and a pharmaceutically acceptable carrier.
10 . A method for preparing a pharmaceutical composition comprising combining a compound prepared according to the process of claim 2 and a pharmaceutically acceptable carrier.
11 . A method for preparing a pharmaceutical composition comprising combining a compound prepared according to the process of claim 3 and a pharmaceutically acceptable carrier.
12 . A method for preparing a pharmaceutical composition comprising combining a compound prepared according to the process of claim 4 and a pharmaceutically acceptable carrier.
