CANINE IgG NUCLEIC ACID MOLECULES
Abstract
The invention relates to canine immunoglobulin G (IgG) and canine interleukin-13 receptors (IL-13R) as well as fusion proteins containing canine IgG and/or canine IL-13R. In particular, the present invention discloses nucleic acid molecules encoding canine IgG, including species-specific regions of the heavy chain of canine IgG, and canine IL-13R alpha chain (IL-13Rα) proteins, particularly canine interleuken receptor alpha 1 (IL-13Rα1) and canine interleuken receptor alpha 2 (IL-13Rα2) proteins. Also included are canine IgG and IL-13Rα proteins, antibodies having selectivity for such proteins, inhibitors of such proteins and/or nucleic acid molecules, cells transformed with said nucleic acid molecules, assays employing such cells, nucleic acids molecules, proteins, antibodies and/or inhibitors, and therapeutic compositions comprising said nucleic acids molecules, proteins, antibodies and/or inhibitors. Also included are kits containing said molecules or chimera thereof, including their use to evaluate and regulate an immune response in an animal.

60 . An isolated nucleic acid molecule selected from the group consisting of:
(a) an isolated nucleic acid molecule comprising a nucleotide sequence that encodes at least a portion of a canine immunoglobulin G (IgG) protein, wherein said portion comprises at least one region selected from the group consisting of a CH1 region, a CH2 region, a CH3 region, a hinge region, an IgG receptor binding region, and a complement component C3 binding region; and (b) an isolated nucleic acid molecule fully complementary in sequence to the nucleic acid molecule of (a).
61 . The isolated nucleic acid molecule of claim 60 , wherein said portion comprises more than one region selected from the group consisting of a CH1 region, a CH2 region, a CH3 region, a hinge region, an IgG receptor binding region, and a complement component C3 binding region.
62 . The isolated nucleic acid molecule of claim 60 , wherein said hinge region comprises an amino acid sequence at least 95% identical over its entire length to a sequence selected from the group consisting of SEQ ID NO:2, SEQ ID NO:8, SEQ ID NO:14 and SEQ ID NO:23.
63 . The isolated nucleic acid molecule of claim 60 , wherein said hinge region comprises an amino acid sequence selected from the group consisting of SEQ ID NO:2, SEQ ID NO:8, SEQ ID NO:14 and SEQ ID NO:23.
64 . The isolated nucleic acid molecule of claim 60 , wherein said CH1 region comprises an amino acid sequence at least 95% identical to a sequence selected from the group consisting of:
(a) the sequence consisting of amino acid residues 138-235 from SEQ ID NO:5; (b) the sequence consisting of amino acid residues 139-236 from SEQ ID NO:20; (c) the sequence consisting of amino acid residues 140-237 from SEQ ID NO:11; and (d) the sequence consisting of amino acid residues 142-239 from SEQ ID NO:17.
65 . The isolated nucleic acid molecule of claim 60 , wherein said CH2 region comprises an amino acid sequence at least 95% identical to a sequence selected from the group consisting of:
(a) the sequence consisting of amino acid residues 253-358 from SEQ ID NO:5; (b) the sequence consisting of amino acid residues 259-364 from SEQ ID NO:20; (c) the sequence consisting of amino acid residues 255-360 from SEQ ID NO:11; and (d) the sequence consisting of amino acid residues 260-365 from SEQ ID NO:17.
66 . The isolated nucleic acid molecule of claim 60 , wherein said CH3 region comprises an amino acid sequence at least 95% identical to a sequence selected from the group consisting of:
(a) the sequence consisting of amino acid residues 359-468 from SEQ ID NO:5; (b) the sequence consisting of amino acid residues 365-473 from SEQ ID NO:20; (c) the sequence consisting of amino acid residues 361-470 from SEQ ID NO:11; and (d) the sequence consisting of amino acid residues 366-474 from SEQ ID NO:17.
67 . The isolated nucleic acid molecule of claim 60 , wherein said canine IgG protein comprises an amino acid sequence at least 95% identical over it's entire length to a sequence selected from the group consisting of SEQ ID NO:5, SEQ ID NO:11, SEQ ID NO:17, and SEQ ID NO:20.
68 . The isolated nucleic acid molecule of claim 60 , wherein said portion has IgE receptor binding activity, complement component C3 binding activity, or is capable of eliciting an immune response to a canine IgG protein.
69 . The isolated nucleic acid molecule of claim 60 , wherein said nucleotide sequence encodes a protein at least 95% identical to SEQ ID NO:5, SEQ ID NO:11, SEQ ID NO:17 or SEQ ID NO:21.
70 . The isolated nucleic acid molecule of claim 60 , wherein said nucleotide sequence encodes a protein comprising SEQ ID NO:5, SEQ ID NO:11, SEQ ID NO 17 or SEQ ID NO:21.
71 . An isolated nucleic acid molecule selected from the group consisting of:
(a) an isolated nucleic acid molecule comprising at least a portion of a nucleotide sequence that is at least 95% identical over its entire length to a polynucleotide sequence selected from the group consisting of SEQ ID NO:4, SEQ ID NO:10, SEQ ID NO:16 and SEQ ID NO:20, wherein said portion encodes at least one canine immunoglobulin G (IgG) protein region selected from the group consisting of a CH1 region, a CH2 region, a CH3 region, a hinge region, an IgG receptor binding region, and a complement component C3 binding region; and (b) an isolated nucleic acid molecule fully complementary in sequence to the nucleic acid molecule of (a).
72 . The isolated nucleic acid molecule of claim 71 , wherein said portion encodes more than one region selected from the group consisting of a CH1 region, a CH2 region, a CH3 region, a hinge region, an IgG receptor binding region, and a complement component C3 binding region.
73 . The isolated nucleic acid molecule of claim 71 , wherein said hinge region is encoded by a nucleic acid sequence at least 95% identical to a sequence selected from the group consisting of SEQ ID NO:1, SEQ ID NO:7, SEQ ID NO:13 and SEQ ID NO:22.
74 . The isolated nucleic acid molecule of claim 71 , wherein said CH1 region is encoded by a nucleic acid sequence at least 95% identical to a sequence selected from the group consisting of:
(a) the sequence consisting of nucleotides 481-774 from SEQ ID NO:4; (b) the sequence consisting of nucleotides 446-739 from SEQ ID NO:19; (c) the sequence consisting of nucleotides 465-758 from SEQ ID NO:10; and (d) the sequence consisting of nucleotides 455-748 from SEQ ID NO:16.
75 . The isolated nucleic acid molecule of claim 71 , wherein said CH2 region comprises an amino acid sequence at least 95% identical to a sequence selected from the group consisting of:
(a) the sequence consisting of nucleotides 826-1143 from SEQ ID NO:4; (b) the sequence consisting of nucleotides 806-1123 from SEQ ID NO:19; (c) the sequence consisting of nucleotides 810-1127 from SEQ ID NO:10; and (d) the sequence consisting of nucleotides 809-1126 from SEQ ID NO:16.
76 . The isolated nucleic acid molecule of claim 71 , wherein said CH3 region comprises an amino acid sequence at least 95% identical to a sequence selected from the group consisting of:
(a) the sequence consisting of amino acid residues 1144-1473 from SEQ ID NO:4; (b) the sequence consisting of amino acid residues 1124-1450 from SEQ ID NO:19; (c) the sequence consisting of amino acid residues 1128-1457 from SEQ ID NO:10; and (d) the sequence consisting of amino acid residues 1127-1453 from SEQ ID NO:16.
77 . The isolated nucleic acid molecule of claim 71 , wherein said portion encodes a protein that has IgE receptor binding activity, complement component C3 binding activity, or is capable of eliciting an immune response to a canine IgG protein.
78 . The isolated protein of claim 71 , wherein said nucleic acid molecule comprises a nucleic acid sequence at least 95% identical to SEQ ID NO:4, SEQ ID NO:10, SEQ ID NO:16 or SEQ ID NO:20.
