Abstract
A trace amine receptor 1 has been isolated from the genome of Cercopithecus aethiops (African green monkey). Described is the amino acid sequence of the receptor, the nucleic acid encoding the receptor, and methods for using the receptor to identify analytes that are agonists or antagonists of the receptor.

23 . A nucleic acid molecule comprising a sequence of nucleotides that encodes the isolated African green monkey trace amine receptor 1 (agm TA1 receptor) protein set forth in SEQ ID NO:2.
24 . The nucleic acid molecule of claim 23 wherein the sequence of nucleotides is SEQ ID NO:1.
25 . A vector comprising an agm TA1 receptor expression cassette comprising a transcriptional initiation region in a host cell, the nucleic acid molecule of claim 24 operably linked to the transcriptional initiation region, and a transcriptional termination region functional in the host cell.
26 . A host cell comprising the agm TA1 receptor expression cassette of claim 25 as part of an extrachromosomal element or integrated into the genome of a host cell.
27 . A method of making the agm TA1 receptor, the method comprising culturing a host cell of claim 26 and recovering said agm TA1 receptor.
28 . A method for identifying an analyte which binds to the agm TA1 receptor of claim 23 comprising:
(a) providing a host cell which produces the agm TA1 receptor; (b) incubating the host cell in a medium with the analyte; and (c) determining the amount of analyte bound to the host cell.
29 . A method for identifying an analyte that modulates the activity of the agmTA1 receptor of claim 23 comprising:
(a) providing a host cell which produces the agmTA1 receptor; (b) incubating the host cell in a medium with the analyte; and (c) determining the activity of the agmTA1 receptor wherein a change in the activity of the agmTA1 receptor indicates the analyte modulates activity of the agmTA1 receptor.
30 . The method of claim 29 wherein the activity of the agm TA1 receptor is determined by measuring a change in the intracellular concentration of Ca 2+ in the presence of the analyte.
31 . The method of claim 29 wherein the activity of the agm TA1 receptor is determined by measuring a change in the intracellular concentration of cyclic adenosine monophosphate (cAMP).
32 . The method of claim 29 wherein the activity of the agmTA1 receptor is determined by measuring a change in the activity of protein kinase A.
33 . A method for determining whether an analyte is an agmTA1 receptor agonist comprising:
(a) providing a membrane which has the agmTA1 receptor protein of SEQ ID NO: 2 integrated therein; (b) incubating the membrane in the presence of the analyte and labeled GTP for a time sufficient for the labeled GTP to be associated with the membrane when the analyte is an agonist; and (c) separating the membrane from unbound labeled GTP and determining the amount of labeled GTP associated with the membrane wherein presence of the labeled GTP associated with the membrane indicates that the analyte is an agonist.Join the waitlist — get patent alerts
