Claims

Abstract

Compositions and methods for regulating genes of interest are provided. The compositions comprise sense-antisense double stranded nucleic acid molecules, which have homology to a target nucleic acid sequence in a cell. The compositions and methods are useful for the treatment of disease and for preventing the proliferation of neoplastic cells.

A method comprising introducing a double stranded nucleic acid molecule into a cell, wherein said double stranded nucleic acid molecule comprises a first strand comprising an RNA sequence and a second strand comprising a polynucleotide bound to said RNA sequence, further wherein said polynucleotide shares up to 100% homology to said first strand and to a target molecule.  
     
     
         14 . The method of  claim 13 , wherein said polynucleotide is RNA or DNA.  
     
     
         15 . The method of  claim 14 , wherein said polynucleotide is DNA.  
     
     
         16 . The method of  claim 14 , wherein said polynucleotide is RNA.  
     
     
         17 . The method of  claim 14 , wherein said polynucleotide comprises modified phosphates.  
     
     
         18 . The method of  claim 14 , wherein said polynucleotide is a sequence of from 10 to 200 nucleotides.  
     
     
         19 . The method of  claim 14 , wherein said first strand and said second strand comprise mismatched nucleotides.  
     
     
         20 . The method of  claim 14 , wherein said first strand or said second strand comprises an unhybridized overhang region.  
     
     
         21 . The method of  claim 14 , wherein said double stranded nucleic acid molecule comprises a stem-loop structure.  
     
     
         22 . The method of  claim 14 , wherein the target molecule is an RNA selected from the group consisting of a viral RNA, an mRNA, and a non-coding RNA.

A cell produced by the process of introducing a double stranded nucleic acid molecule into a target cell, wherein said double stranded nucleic acid molecule comprises a first strand comprises an RNA sequence and a second strand comprises a polynucleotide bound to said RNA sequence, further wherein said polynucleotide shares up to 100% homology to said first strand and to a target molecule.
