Abstract

Disclosed is a method for detecting nucleic acid hybridization by using intercalator binding to hybridized nucleic acid, wherein oxidation-reduction of transition metallic complex is induced to cause electrochemiluminescence, thereby providing a method for detecting nucleic acid hybridization without a special labeling.

A method for detecting nucleic acid hybridization, comprising steps of using nucleic acid intercalator under proper applied voltage to induce an electrochemiluminescence reaction of transition metallic complex; and detecting the quantity of light generated thereby, wherein the intercalator peculiarly binds to double-stranded nucleic acid which is formed by hybridization between probe nucleic acid fixed on a metal surface and target nucleic acid in sample solution and causes an electrochemiluminescence reaction by inducing an oxidation-reduction reaction of transition metallic complex.  
     
     
         2 . The method for detecting nucleic acid hybridization of  claim 1 , wherein the intercalator is selected from a group including doxorubicin, daunorubicin and DAPI (4′,6-Diamidino-2-phenylindole).  
     
     
         3 . The method for detecting nucleic acid hybridization of  claim 1 , wherein the transition metallic complex is [Ru(bpy) 3   2+ ] or [Ru(phen) 3   2+ ].  
     
     
         4 . The method for detecting nucleic acid hybridization of  claim 1 , wherein an applied voltage ranges from +1.12 to +1.20 V.  
     
     
         5 . The method for detecting nucleic acid hybridization of  claim 1 , wherein the nucleic acid is selected from a group including oligonucleotide, DNA, RNA, PNA and cDNA.  
     
     
         6 . DAPI (4′,6-Diamidino-2-phenylindole) used as an efficient intercalator for detecting a result of nucleic acid hybridization.
