Use of parvovirus capsid particles in the inhibition of cell proliferation and migration
Abstract
The invention described herein relates to the discovery of methods and compositions for the inhibition of growth and/or migration of cells that have a receptor that interacts with a parvovirus B19 capsid or fragment thereof (e.g., a P antigen containing cell), including but not limited to, cells of hematopoietic origin and endothelial cells. More specifically, parvovirus capsid particles or fragments of parvovirus capsid proteins are used to manufacture medicaments that can be administered to a subject to inhibit hematopoietic progenitor cell growth (e.g., prior to stem cell transplantation), endothelial cell growth, (e.g., as an anti-tumorigenesis treatment or to prevent restenosis or fibrotic build up following prosthetic implantation), or to prevent disorders that involve the abnormal proliferation of cells that have the P antigen (e.g., Polycythemia Vera).
Claims
exact text as granted — not AI-modified1 . A method of inhibiting hematopoiesis in a subject in need thereof comprising:
identifying a subject in need of an inhibition of hematopoiesis; and providing to said subject an amount of a fragment of parvovirus B19 major capsid protein (VP2) sufficient to inhibit hematopoiesis, wherein said fragment is at least three amino acids in length, and whereby hematopoiesis in said subject is inhibited.
2 . The method of claim 1 , wherein said subject in need is identified as having a hematological proliferative disorder.
3 . The method of claim 2 , wherein said hematological proliferative disorder is polycythemia vera.
4 . The method of claim 1 , wherein said fragment is at least 20 amino acids in length.
5 . The method of claim 1 , wherein said fragment comprises the sequence glutamine-glutamine-tyrosine.
6 . The method of claim 1 , wherein said fragment comprises the sequence of SEQ. ID. No. 5.
7 . The method of claim 1 , further comprising measuring the inhibition of hematopoeisis.
8 . A method of inhibiting hematopoiesis in a subject in need thereof comprising:
identifying a subject in need of an inhibition of hematopoiesis; and providing to said subject an amount of a fragment of parvovirus B19 major capsid protein (VP2) sufficient to inhibit hematopoiesis, and whereby hematopoiesis in said subject is inhibited.
9 . The method of claim 8 , wherein said subject in need is identified as having a hematological proliferative disorder.
10 . The method of claim 9 , wherein said hematological proliferative disorder is polycythemia vera.
11 . The method of claim 8 , wherein said fragment is at least 20 amino acids in length.
12 . The method of claim 8 , wherein said fragment comprises the sequence glutamine-glutamine-tyrosine.
13 . The method of claim 8 , wherein said fragment comprises the sequence of SEQ. ID. No. 5.
14 . The method of claim 8 , further comprising measuring the inhibition of hematopoeisis.
15 . A method of inhibiting endothelial cell proliferation comprising:
identifying a subject in need of an inhibition of endothelial cell proliferation; and providing to said subject a parvovirus B19 major capsid protein (VP2) or a fragment thereof in an amount sufficient to inhibit the proliferation of said endothelial cells, whereby said proliferation is inhibited.
16 . The method of claim 15 , wherein parvovirus B19 major capsid protein (VP2) is provided.
17 . The method of claim 15 , wherein said fragment is at least 20 amino acids in length.
