Abstract

The invention relates to an isolated polynucleotide having a polynucleotide sequence which codes for the cstA gene, and a host-vector system having a coryneform host bacterium in which the cstA gene is present in attenuated form and a vector which carries at least the cstA gene according to SEQ ID No 1, and the use of polynucleotides which comprise the sequences according to the invention as hybridization probes.

         31 . A method for the fermentative production of L-amino acids, comprising: 
 a) culturing coryneform bacteria, which include an overexpressed cstA gene comprising the polynucleotide sequence of SEQ ID NO: 1, in a medium suitable for the expression of the cstA gene to thereby produce L-amino acids.    
     
     
         32 . The method according to  claim 31 , further comprising: 
 b) isolating the L-amino acids.    
     
     
         33 . The method as claimed in  claim 31 , wherein the L-amino acid produced is L-lysine.  
     
     
         34 . The method as claimed in  claim 31 , wherein the bacteria comprise, at the same time, one or more endogenous  Corynebacterium glutamicum  genes which are overexpressed, wherein the one or more genes is/are selected from the group consisting of: 
 the gene which encodes feed back resistant aspartate kinase,    the gene which encodes dihydrodipicolinate synthase,    the gene which encodes glycerolaldehyde 3-phosphate dehydrogenase,    the gene which encodes 3-phosphoglycerate kinase,    the gene which encodes pyruyate carboxylase,    the gene which encodes triose phosphate isomerase,    the gene which encodes a protein that exports lysine, and    the gene which encodes the Zwa1 protein.    
     
     
         35 . The method as claimed in  claim 31 , wherein the bacteria comprise, at the same time, one or more endogenous  Corynebacterium glutamicum  genes which are eliminated, wherein said one or more genes is/are selected from the group consisting of: 
 the gene which encodes phosphoenol pyruvate carboxykinase,    the gene which encodes glucose 6-phosphate isomerase,    the gene which encodes pyruvate oxidase, and    the gene which encodes the Zwa2 protein.    
     
     
         36 . The method according to  claim 31 , wherein the bacteria are  Corynebacterium glutamicum.    
     
     
         37 . The method according to  claim 31 , wherein the cstA gene is overexpressed by increasing the copy number of the cstA gene or by operably linking a promoter to the cstA gene.  
     
     
         38 . A method for the fermentative production of L-amino acids, comprising: 
 a) culturing coryneform bacteria, which include an overexpressed cstA gene having a polynucleotide sequence which encodes the polypeptide comprising the amino acid sequence of SEQ ID NO: 2, in a medium suitable for the expression of the cstA gene to thereby produce L-amino acids.    
     
     
         39 . The method according to  claim 38 , further comprising: 
 b) isolating the L-amino acids.    
     
     
         40 . The method as claimed in  claim 38 , wherein the L-amino acid produced is L-lysine.  
     
     
         41 . The method as claimed in  claim 38 , wherein the bacteria comprise, at the same time, one or more endogenous  Corynebacterium glutamicum  genes which are overexpressed, wherein the one or more genes is/are selected from the group consisting of:

         42 . The method as claimed in  claim 38 , wherein the bacteria comprise, at the same time, one or more endogenous  Corynebacterium glutamicum  genes which are eliminated, wherein said one or more genes is/are selected from the group consisting of: 
 the gene which encodes phosphoenol pyruvate carboxykinase,    the gene which encodes glucose 6-phosphate isomerase,    the gene which encodes pyruvate oxidase, and    the gene which encodes the Zwa2 protein.    
     
     
         43 . The method according to  claim 38 , wherein the bacteria are  Corynebacterium glutamicum.    
     
     
         44 . The method according to  claim 38 , wherein the cstA gene is overexpressed by increasing the copy number of the cstA gene or by operably linking a promoter to the cstA gene.  
     
     
         45 . A method for the fermentative production of L-amino acids, comprising: 
 a) culturing coryneform bacteria, which include an overexpressed cstA gene having a polynucleotide sequence that comprises nucleotides 200 to 2515 of SEQ ID NO: 1, in a medium suitable for the expression of the cstA gene to thereby produce L-amino acids,    wherein the cstA gene is overexpressed by increasing the copy number of the cstA gene or by operably linking a promoter to the cstA gene.    
     
     
         46 . The method according to  claim 45 , further comprising: 
 b) isolating the L-amino acids.    
     
     
         47 . The method as claimed in  claim 45 , wherein the L-amino acid produced is L-lysine.  
     
     
         48 . The method as claimed in  claim 45 , wherein the bacteria comprise, at the same time, one or more endogenous  Corynebacterium glutamicum  genes which are overexpressed, wherein the one or more genes is/are selected from the group consisting of: 
 the gene which encodes feed back resistant aspartate kinase,    the gene which encodes dihydrodipicolinate synthase,    the gene which encodes glycerolaldehyde 3-phosphate dehydrogenase,    the gene which encodes 3-phosphoglycerate kinase,    the gene which encodes pyruvate carboxylase,    the gene which encodes triose phosphate isomerase,    the gene which encodes a protein that exports lysine, and    the gene which encodes the Zwa1 protein.    
     
     
         49 . The method as claimed in  claim 45 , wherein the bacteria comprise, at the same time, one or more endogenous  Corynebacterium glutamicum  genes which are eliminated, wherein said one or more genes is/are selected from the group consisting of: 
 the gene which encodes phosphoenol pyruvate carboxykinase,    the gene which encodes glucose 6-phosphate isomerase,    the gene which encodes pyruvate oxidase, and    the gene which encodes the Zwa2 protein.    
     
     
         50 . A method for the fermentative production of L-amino acids, comprising: 
 a) culturing coryneform bacteria, which include an overexpressed cstA gene having a polynucleotide sequence that comprises a fragment of SEQ ID NO: 1 which encodes the amino acid sequence of SEQ ID NO: 2, in a medium suitable for the expression of the cstA gene to thereby produce L-amino acids,    wherein the cstA gene is overexpressed by increasing the copy number of the cstA gene or by operably linking a promoter to the cstA gene.    
     
     
         51 . The method according to  claim 50 , further comprising: 
 b) isolating the L-amino acids.    
     
     
         52 . The method as claimed in  claim 50 , wherein the L-amino acid produced is L-lysine.  
     
     
         53 . The method as claimed in  claim 50 , wherein the bacteria comprise, at the same time, one or more endogenous  Corynebacterium glutamicum  genes which are overexpressed, wherein the one or more genes is/are selected from the group consisting of: 
 the gene which encodes feed back resistant aspartate kinase,    the gene which encodes dihydrodipicolinate synthase,    the gene which encodes glycerolaldehyde 3-phosphate dehydrogenase,    the gene which encodes 3-phosphoglycerate kinase,    the gene which encodes pyruvate carboxylase,    the gene which encodes triose phosphate isomerase,    the gene which encodes a protein that exports lysine, and    the gene which encodes the Zwa1 protein.    
     
     
         54 . The method as claimed in  claim 50 , wherein the bacteria comprise, at the same time, one or more endogenous  Corynebacterium glutamicum  genes which are eliminated, wherein said one or more genes is/are selected from the group consisting of: 
 the gene which encodes phosphoenol pyruvate carboxykinase,    the gene which encodes glucose 6-phosphate isomerase,    the gene which encodes pyruvate oxidase, and    the gene which encodes the Zwa2 protein.
