Abstract
The present invention includes recombinant proteins derived from Clostridium botulinum toxins. In particular, soluble recombinant Clostridium botulinum type A, type B and type E toxin proteins are provided. Methods which allow for the isolation of recombinant proteins free of significant endotoxin contamination are provided. The soluble, endotoxin-free recombinant proteins are used as immunogens for the production of vaccines and antitoxins. These vaccines and antitoxins are useful in the treatment of humans and other animals at risk of intoxication with clostridial toxin.

25 . A soluble recombinant botulinum toxin, comprising the Clostridium botulinum a toxin amino acid sequence of SEQ ID NO:23, the toxin being obtained from an aerobic host cell by a process of recombinantly expressing the toxin using the aerobic host cell, the toxin being soluble.
26 . The soluble recombinant botulinum toxin of claim 25 , further comprising a poly-histidine tract.
27 . The soluble recombinant botulinum toxin of claim 25 , wherein the toxin comprises the amino acid sequence of SEQ ID NO:23.
28 . The soluble recombinant botulinum toxin of claim 25 , wherein the toxin is obtained from E. Coli bacteria.
29 . The soluble recombinant botulinum toxin of claim 25 , wherein the toxin is soluble in an aqueous environment.
30 . The soluble recombinant botulinum toxin of claim 25 , wherein the toxin exists in solution in the cytoplasm of the host cell.
31 . The soluble recombinant botulinum toxin of claim 25 , wherein the toxin is soluble in a solution free of significant amounts of at least one of ionic detergents and denaturants.
