Claims

Abstract

Phenyl acetamide compounds are described, including compounds of Formula I: or a solvate, hydrate or pharmaceutically acceptable salt thereof; wherein R 3 -R 6 , R 11 , B, Y and W are set forth in the specification. The compounds of the invention are potent inhibitors of proteases, especially trypsin-like serine proteases, such as thrombin and factor Xa. Compositions for inhibiting loss of blood platelets, inhibiting formation of blood platelet aggregates, inhibiting formation of fibrin, inhibiting thrombus formation, and inhibiting embolus formation are described. Other uses of compounds of the invention are as anticoagulants either embedded in or physically linked to materials used in the manufacture of devices used in blood collection, blood circulation, and blood storage, such as catheters, blood dialysis machines, blood collection syringes and tubes, blood lines and stents. Additionally, the compounds can be detectably labeled and employed for in vivo imaging of thrombi.

 (R 2 ) 2 CH(CH 2 ) r , where r is 0-4 and each R 2  can be the same or different, and wherein (R 2 ) 2  can also form a ring with CH represented by C 3-9  cycloalkyl, C 7-12  bicylic alkyl, C 10-16  tricylic alkyl, or a 5- to 7-membered mono- or bicyclic heterocyclic ring which can be saturated or unsaturated, and which contains from one to three heteroatoms selected from the group consisting of N, O and S,

 a 5- to 7-membered mono- or a 9- to 10-membered bicyclic heterocyclic ring or non-heterocyclic ring which can be saturated or unsaturated, wherein the heterocyclic ring contains from one to four heteroatoms selected from the group consisting of N, O and S, and wherein the heterocyclic or non-heterocyclic ring is unsubstituted or substituted with halogen or hydroxy,

 R 3 , R 4 , R 5  and R 6  are independently hydrogen, C 1-12  alkyl, C 3-9  cycloalkyl, halogen, C 2-20  alkenyl, C 2-20  alkynyl, optionally substituted C 6-14  aryl, optionally substituted C 6-14 ar(C 1-12 )alkyl, optionally substituted heteroaryl, halo(C 1-12 )alkyl, C 1-12  alkoxy, C 6-14  aryloxy, heteroaryloxy, halo(C 1-20 )alkoxy or hydroxy(C 1-12 )alkyl;    R 11  is hydrogen, C 1-12  alkyl or C 2-20  alkenyl;    R 7 , R 8 , R 9  and R 10  are independently hydrogen, C 1-12  alkyl, C 6-14 ar(C 1-12 )alkyl, C 6-14 aryl, hydroxy(C 1-12 )alkyl, amino(C 1-12 )alkyl, mono(C 1-12 )alkylamino(C 1-12 )alkyl, di(C 1-12 )alkylamino(C 1-12 )alkyl, or carboxy(C 1-12 )alkyl;    R 18  is C 1-12  alkyl, C 3-9  cycloalkyl or C 6-14  aryl, each of which is optionally substituted with amino, mono(C 1-12 )alkylamino, di(C 1-12 )alkylamino, C 1-20  alkoxy, hydroxy, carboxy, C 1-20 alkoxycarbonyl, C 6-14  aryloxycarbonyl, C 6-14 ar(C 1-20 )alkoxycarbonyl, C 6-14  aryl, C 5-10  heteroaryl, acylamino, cyano or trifluoromethyl;    R a , R b  and R c  are independently C 1-12  alkyl, C 1-20  alkoxy, C 6-14  aryloxy, C 6-14 ar(C 1-20 )alkoxy, or C 1-20  alkoxycarbonyloxy;    R 13  is C 1-12  alkyl, C 1-20  alkoxy, C 6-14  aryloxy or C 1-20  alkoxycarbonyl;    R 14  and R 15  are independently C 1-12  alkyl, C 3-9  cycloalkyl or C 1-20  alkoxy; and    R 16  and R 17  are independently C 1-12  alkyl, C 1-20  alkoxy, C 6-14  aryloxy or C 1-20  alkoxycarbonyl.

         7 . A compound according to  claim 1 , wherein W is R 1 , where R 1  is R 2  or R 2 CF 2 C(R 12 ) 2 (CH 2 ) q , and R 2  is either optionally substituted aryl or C 1-7  alkyl substituted with aryl; R 12  is hydrogen; and q is zero.  
     
     
         8 . A compound according to  claim 1 , wherein R 6  is C 1-6  alkyl or halogen.  
     
     
         9 . A compound according to  claim 8 , wherein R 6  is methyl, chloro or fluoro.  
     
     
         10 . A compound according to  claim 9 , wherein R 6  is chloro while R 3  is fluoro or hydroxy.  
     
     
         11 . A compound according to  claim 1 , wherein R 11  is hydrogen.  
     
     
         12 . A compound according to  claim 1 , wherein R a , R b , R c  and R 13  are each hydrogen.  
     
     
         13 . A compound according to  claim 1 , wherein each of R 7 , R 8 , R 9  and R 10  are hydrogen.

         37 . A method of inhibiting the action of a proteolytic enzyme, comprising contacting said enzyme with a compound of  claim 4 .
