Method for isolating terpene trilactones (ginkgolides, bilobadide) from leaves and pharmaceutical powders of ginkgo biloba
Abstract
A method of isolating terpene trilactones from Ginkgo biloba plant material or extract comprising the steps of suspending the plant material or extract in either water or an aqueous solution of an oxidation reagent; extracting the terpene trilactones using an acceptable extraction agent; separating the organic layer from the aqueous layer; washing the organic layer with an acceptable aqueous salt or hydroxide solution, which may be an alkaline solution; and drying the organic layer to form a dried extract containing terpene trilactones. Further purification by treatment with or filtration over activated charcoal, by treatment with or filtration over alumina and by recrystallization with an acceptable solvent or solvent mixture leads to extracts with a content of terpene trilactones higher than 50%. Unwanted levels of ginkgolic acids are reduced to acceptable levels by reversed phase chromatography.

72 . A composition comprising isolates from Ginkgo Biloba plant material or extract made by a process comprising the steps of:
(1) suspending Ginkgo Biloba plant material or extract in the presence of an oxidation reagent, (2) extracting terpene trilactones from the plant material or extract using an acceptable extraction agent, and (3) separating a resulting organic layer from a resulting aqueous layer to thereby isolate the terpene trilactones in the organic layer, so as to thereby make the composition.
73 . The composition of claim 72 , wherein in the process the oxidation reagent is in aqueous solution.
74 . The composition of claim 72 , wherein in the process the oxidation reagent is hydrogen peroxide.
75 . The composition of claim 72 , wherein in the process the aqueous solution contains 0.1% to 50% oxidation reagent.
76 . The composition of claim 72 , wherein in the process the aqueous solution further comprises an acid.
77 . The composition of claim 72 , wherein in the process the aqueous solution contains 0.1% to 15% acid.
78 . The composition of claim 72 , wherein in the process the acid is selected from the group consisting of acetic acid, hydrochloric acid, nitric acid, phosphoric acid and sulfuric acid.
79 . The composition of claim 72 , wherein in the process the acceptable extraction agent is selected from the group consisting of lower acetates, lower ketones, lower ether, lower alcohols and benzenes.
80 . The composition of claim 72 , wherein the process further comprises at least a first washing step to wash the organic layer with an acceptable aqueous salt or hydroxide solution.
81 . The composition of claim 80 , wherein in the process the acceptable aqueous solution is a solution of a salt or hydroxide selected from the group consisting of ammonium chloride, sodium carbonate, sodium bicarbonate, potassium carbonate, sodium hydroxide, potassium hydroxide, sodium thiosulfate, sodium sulfite and sodium hydrosulfide.
82 . The composition of claim 81 , wherein in the process the acceptable aqueous salt or hydroxide solution is a solution of a salt selected from the group consisting of sodium thiosulfate, sodium sulfite and sodium hydrosulfide.
83 . The composition of claim 80 , wherein in the process the acceptable aqueous salt solution is an aqueous alkali salt solution.
84 . The composition of claim 83 , wherein in the process the aqueous alkali salt solution has a pH of between about 7.5 and 9.5.
85 . The composition of claim 82 , wherein in the process the aqueous hydroxide solution is selected from the group consisting of a sodium hydroxide solution and a potassium hydroxide solution.
86 . The composition of claim 80 , wherein the process further comprises a second washing step to wash the organic layer with an acceptable aqueous salt or hydroxide solution.
87 . The composition of claim 86 , wherein in the process the acceptable aqueous salt or hydroxide solutions of the second wash step is selected from the group consisting of ammonium chloride, sodium carbonate, sodium bicarbonate, potassium carbonate, sodium hydroxide, potassium hydroxide, sodium thiosulfate, sodium sulfite and sodium hydrosulfide.
88 . The composition of claim 86 , wherein in the process the second washing step is performed with a solution that is different from the solution used in the first washing step.
89 . The composition of claim 72 , wherein the process further comprises a step to destroy excess oxidation reagent in the organic layer by contacting the organic layer with a metal or a nonmetal catalyst.
90 . The composition of claim 72 , wherein the process further comprises a step of drying the organic layer to form an extract containing terpene trilactones.
