Abstract

Novel receptor polypeptides, polynucleotides encoding the polypeptides, and related compositions and methods are disclosed. The polypeptides comprise an extracellular domain of a cell-surface receptor that is expressed in pancreas, small intestine, colon and thymus. The polypeptides may be used within methods for detecting ligands that promote the proliferation and/or differentiation of these organs.

1 . An isolated polynucleotide encoding a cytokine receptor polypeptide, the polynucleotide comprising from amino acid residues 18 to 228 of SEQ ID NO:2.  
     
     
         2 . The isolated polynucleotide of  claim 1 , wherein the polynucleotide comprises from nucleotides 85 to 717 of SEQ ID NO:1.  
     
     
         3 . The isolated polynucleotide of  claim 1 , wherein the polynucleotide encodes a polypeptide comprising the amino acid sequence of SEQ ID NO:2.  
     
     
         4 . The isolated polynucleotide of  claim 1 , wherein the polynucleotide comprises SEQ ID NO:1.  
     
     
         5 . The isolated polynucleotide of  claim 1 , wherein the encoded polypeptide further comprises a transmembrane domain.  
     
     
         6 . The isolated polynucleotide of  claim 5 , wherein the transmembrane domain comprises amino acid residues 229 to 251 of SEQ ID NO:2.  
     
     
         7 . The isolated polynucleotide of  claim 1 , wherein the encoded polypeptide further comprises an intracellular domain.  
     
     
         8 . The isolated polynucleotide of  claim 7 , wherein the intracellular domain comprises amino acid residues 252 to 574 of SEQ ID NO:2.  
     
     
         9 . The isolated polynucleotide of  claim 1 , wherein the encoded polypeptide further comprises an affinity tag.  
     
     
         10 . The isolated polynucleotide of  claim 9 , wherein the affinity tag is selected from the group consisting of polyhistidine, protein A, glutathione S transferase, substance P, and an immunoglobulin heavy chain constant region.

An isolated polynucleotide encoding a polypeptide selected from the group consisting of amino acid residues 1 to 228 of SEQ ID NO:2, amino acid residues 1 to 251 of SEQ ID NO:2, amino acid residues 1 to 574 of SEQ ID NO:2, amino acid residues 2 to 228 of SEQ ID NO:2, amino acid residues 2 to 251 of SEQ ID NO:2, amino acid residues 2 to 574 of SEQ ID NO:2, amino acid residues 229 to 251 of SEQ ID NO:2, amino acid residues 229 to 574 of SEQ ID NO:2 and amino acid residues 252 to 574 of SEQ ID NO:2.  
     
     
         12 . An expression vector comprising the following operably linked elements: 
 a transcription promoter;    a DNA segment encoding a cytokine receptor polypeptide, wherein the polypeptide comprises amino acid residues 18 to 228 of SEQ ID NO:2; and    a transcription terminator.    
     
     
         13 . The expression vector of  claim 12 , wherein the encoded polypeptide further comprises a signal sequence.  
     
     
         14 . The expression vector of  claim 12 , wherein the encoded polypeptide further comprises a transmembrane domain.  
     
     
         15 . The expression vector of  claim 14 , wherein the transmembrane domain comprises amino acid residues 229 to 251 of SEQ ID NO:2.  
     
     
         16 . The expression vector of  claim 12 , wherein the encoded polypeptide further comprises an intracellular domain.  
     
     
         17 . The expression vector of  claim 16 , wherein the intracellular domain comprises amino acid residues 252 to 574 of SEQ ID NO:2.  
     
     
         18 . A transformed or transfected cell into which has been introduced an expression vector according to  claim 12 , wherein the cell expresses a cytokine receptor polypeptide.
