Abstract
Methods and materials for inducing anti-tumor responses in melanoma patients are disclosed. These methods and materials involve gp100-derived polypeptides that contain both a helper T-cell epitope and a cytotoxic T-cell epitope.

1 . A polypeptide comprising an HTL epitope and a CTL epitope, wherein said polypeptide is 13 to 30 amino acids long, and wherein the amino acid sequence of said polypeptide is at least 90% identical to a gp100 amino acid sequence.
2 . The polypeptide of claim 1 , wherein said polypeptide comprises an amino acid sequence as set forth in SEQ ID NO:1, 2, 3, 4, 5, 6, 7, 8, 9, 10, 11, 12, 13, 14, 15, or 16.
3 . The polypeptide of claim 1 , wherein said polypeptide binds more than one MHC class II allele.
4 . The polypeptide of claim 1 , wherein said polypeptide binds HLA-DR-53 or HLA-DQw6.
5 . The polypeptide of claim 1 , wherein said CTL epitope comprises MLGTHTMEV.
6 . The polypeptide of claim 1 , wherein said CTL epitope comprises VSNDGPTLI.
7 . The polypeptide of claim 1 , wherein said CTL epitope comprises ALNFPGSQK.
8 . A method for inducing an anti-tumor response in a mammal, said method comprising administering a polypeptide to said mammal, wherein said polypeptide comprises an HTL epitope and a CTL epitope, wherein said polypeptide is 13 to 30 amino acids long, and wherein the amino acid sequence of said polypeptide is at least 90% identical to a gp100 amino acid sequence.
9 . The method of claim 8 , wherein said polypeptide comprises an amino acid sequence as set forth in SEQ ID NO:1, 2, 3, 4, 5, 6, 7, 8, 9, 10, 11, 12, 13, 14, 15, or 16.
10 . The method of claim 8 , wherein said polypeptide binds more than one MHC class II allele.
11 . The method of claim 8 , wherein said polypeptide binds HLA-DR-53 or HLA-DQw6.
12 . The method of claim 8 , wherein said CTL epitope comprises MLGTHTMEV.
