Abstract

Disclosed herein is a process for liquid cultivating Schizophyllum commune Fr. for isolation of β-1,6-branched-β-1,3-glucan and a composition for external application containing β-1,6-branched-β-1,3-glucan as an active ingredient, which can defer skin aging, impart skin whitening effect and cure skin damage effectively.

What is claimed is:  
     
         1 . A process for liquid cultivating  Schizophyllum commune  Fr. for isolation of β-1,6-branched-β-1,3-glucan, which comprises steps of; 
 (1) preparing mycelia of  Schizophyllum commune  Fr.;  
 (2) cultivating the mycelia on a liquid medium with pH of 4.0˜8.5 comprising 1˜10% by weight of glucose, 0.1˜1% by weight of yeast extract, 0.05˜0.5% by weight of (NH 4 ) 2 HPO 4  or (NH 4 ) 2 SO 4 , 0.05˜0.5% by weight of KH 2 PO 4  and 0.005˜0.2% by weight of MgSO 4 .7H 2 O;  
 (3) isolating β-1,6-branched-β-1,3-glucan from the liquid medium; and  
 (4) purifying β-1,6-branched-β-1,3-glucan.  
 
     
     
         2 . The process as claimed in    claim 1   , which further comprises a step of adding activated charcoal into said liquid medium in an amount of 0.1˜5% after the cultivation for 2˜4 days and cultivating for further 1˜4 days.  
     
     
         3 . The process as claimed in    claim 1   , wherein the said liquid medium comprises 1˜3% by weight of glucose, 0.3˜0.5% by weight of yeast extract, 0.05˜0.5% by weight of (NH 4 ) 2 HPO 4  or (NH 4 ) 2 SO 4 , 0.05˜0.5% by weight of KH 2 PO 4  and 0.005˜0.2% by weight of MgSO 4 .7H 2 O and which further comprises a step of adding glucose into said liquid medium in an amount of 1˜10% after the cultivation for 3˜5 days and cultivating for further 1˜3 days.  
     
     
         4 . The method as claimed in    claim 1   , wherein the said liquid medium comprises 1˜3% by weight of glucose, 0.3˜0.5% by weight of yeast extract, 0.05˜0.5% by weight of (NH 4 ) 2 HPO 4  or (NH 4 ) 2 SO 4 , 0.05˜0.5% by weight of KH 2 PO 4  and 0.005˜0.2% by weight of MgSO 4 .7H 2 O and which further comprises a step of adding both 0.1˜5% of activated charcoal and 1˜10% of glucose into said liquid medium after the cultivation for 3˜5 days and cultivating for further 1˜5 days.  
     
     
         5 . A composition for external application contains β-1,6-branched-β-1,3-glucan obtained by a process claimed in any one of 1˜4 in an amount of 0.00001˜10% by weight.  
     
     
         6 . The composition as claimed in    claim 5   , wherein said composition has a cosmetic formulation selected from the group consisting of skin lotion, milk lotion, nutrient toilet water, massage creams, nutrient creams, pack and gel.  
     
     
         7 . The composition as claimed in    claim 5   , wherein said composition has a topicum formulation selected from the group consisting of lotion, ointment, gel, cream, patch and spray.
